Multiple microRNAs (miRNAs) are detected in a microarray format using a novel approach that combines a surface enzyme reaction with nanoparticle-amplified SPR imaging (SPRI). The surface reaction of poly(A) polymerase creates poly(A) tails on miRNAs hybridized onto locked nucleic acid (LNA) microarrays. DNA-modified nanoparticles are then adsorbed onto the poly(A) tails and detected with SPRI. This ultrasensitive nanoparticle-amplified SPRI methodology can be used for miRNA profiling at attomole levels. MicroRNAs (miRNAs) are small RNA molecules (19 to 23mers) that can regulate the expression of genes in plants and animals by binding to the 3′-untranslated region of messenger RNAs.1 Several research groups have studied miRNA gene regulation in processes as diverse as cell proliferation, fat metabolism, and cell differentiation.2,3 The recent surge of interest in miRNAs and the related small interfering RNA (siRNA) gene silencing methodology3 has led to an increased need for the ultrasensitive detection and quantitation of small miRNAs in both solution and surface microarray formats.4-7

Monoactivated derivatives of 1,1-dihydroperoxides undergo an unprecedented base-promoted fragmentation to efficiently generate singlet oxygen (1O2) in anhydrous organic solvents. Singlet molecular oxygen (1O2), an important oxidant in chemistry, biology, and medicine, 1a,b is most commonly generated via photosensitized excitation of ground state (3O2) dioxygen. 1b,2 The discovery that 1O2 is also produced from reaction of H2O2 and HOCl led to the discovery of a number of additional methods for chemical generation.1a,3,4 However, many of these “dark ” oxygenations have significant limitations. Due to the short half-life of 1O2 in aqueous media,5 methods based upon reaction of H2O2 with hypohalites,3 alkaline earth metals, 6 transition metals,7 lanthanides,8 or metalloenzymes9 must typically employ biphasic or emulsion conditions for preparative oxidations.10 Thermal generation of 1O2 from phosphite ozonides,11 silyl hydrotrioxides,12 or arene endoperoxides13 can be conducted in organic solvents but requires preparation of unstable precursors. Porcine endogenous retroviruses (PERVs) represent a risk for xenotransplantation using pig cells or organs since they are integrated in the genome of all pigs and infect human cells in vitro. Recombinants between PERV-A and PERV-C have been described in pigs in vivo and found de novo integrated in the genome of somatic cells, but not in the germ line. To study whether PERV-A/C can infect and have a pathogenic effect in normal pigs, German landrace pigs were inoculated with high-titre PERV-A/C. No provirus integration was found in blood cells or in various tissues, and no antibody production was observed, indicating the absence of infection. Xenotransplantation may help to overcome the shortage of allotransplants. However, the transplantation of cells, tissues or organs from pigs to humans may be associated with transmission of zoonotic porcine microorganisms. Porcine endogenous retroviruses (PERVs) represent a special risk, since they are integrated in the genome of all pigs, are released by normal pig cells and infect human cells in vitro (for review see Ref. [1]). Whereas PERV-A and PERV-B are human/polytropic viruses and are present in the genome of all pigs, PERV-C are ecotropic viruses and are present only in some pigs.
