EFFECT OF THIDIAZURON ON IN VITRO REGENERAnON IN THREE BUCKWHEAT CULTIVARS

by Apoloniusz Berbec, Teresa Doroszewska
"... The frequency of shoot regeneration from callus in two diploid (cvs. Kora and Hruszowska) and one tetraploid (cv. Ernka) buckwheat cultivars was very low on BAP and IAA containing media but was substantially improved when a phenyl urea derivative-thidiazuron (TDZ) was substituted for BAP in the rege ..."

The frequency of shoot regeneration from callus in two diploid (cvs. Kora and Hruszowska) and one tetraploid (cv. Ernka) buckwheat cultivars was very low on BAP and IAA containing media but was substantially improved when a phenyl urea derivative-thidiazuron (TDZ) was substituted for BAP in the regeneration medium. Thidiazuron alone had little effect on buckwheat regeneration and was effective only in combination with IAA. The observed shoot regeneration rates on IAA and TDZ-containing media were from ca. 30 to 50 % of shoot producing calli and from 0.53 to 0.98 shoots per callus fragment. Cv. Kora consistently yielded the most regenerated shoots, the regeneration rates of cvs. Hruszowska and Ernka were similar.

... buckwheat. Suggestions from many scientists were made for using plant cell and tissue culture techniques as a means to overcome breeding barriers in buckwheat (Neskovic et al., 1986; Lachmann, 1991; =-=Bohanec, 1995-=-; Joshi and Bimb, 2001). A number of studies in relation to the potential of inter-specific hybrids to overcome cross-incompatibility have used embryos or ovule culture system. As mentioned by Campbel...

CALLUS FORMATION ANDPL ~ REGENERATION IN ANTHER CULTIJRES OF THREE BUCKWHEAT (FAGOPYRUM ESCULENTUMMoench.) CULTIVARS

Viable shoots were regenerated from anthers of one tetraploid (cv. Emka) and two diploid common buckwheat cultivars (cvs. Kora and Hruszowska) when cultures were maintained in a modified MS liquid medium. Callus development and shoot regeneration was consistently highest in the tetraploid cultivar and different in the two diploid cultivars. Viable shoots were produced by 10 % of the calli of the tetraploid and 4 % and 2.5 % of the calli of the diploids. The most prevalent class of the regenerants were tetraploids, regardless of cultivar. No haploids were found among regenerants derived from the diploid cultivars. Likewise, the tetraploid cultivar yielded no diploid regenerants.

...ther than a cultivated species (Kreft, 1989). Biotechnology, including tissue culture techniques, is looked on as a promising tool that might speed up the breeding effort in buckwheat (Lachman, 1991; =-=Bohanec, 1995-=-). Regeneration in vitro can be a breeding approach in its own right since it is a potential source of somatoclonal variation. The investigations on the use of tissue culture in that species has a rel...
