A bacterial strain having potential to produce thermostable alkaline serine-protease was isolated and identified based 16S rRNA phylogenetic analysis as Bacillus sp. RSP12 (AM930518). Evaluation of alkaline protease production pattern at different fermentation strategies revealed that solid-state fermentation is economic and effective for large scale production. Among different agri-materials, green gram husk supported maximum protease production (35,000 U per gram material). Irrespective of fermentation strategy, fermentation (temperature, particle size, moisture content and pH), nutrition (yeast extract and maltose), and biomaterial related (inoculum size and incubation time) factors play vital role and regulate the enzyme production levels. Significant variation, more than 100 % increase in enzyme production levels (32000-73000 U per gram material) is observed. Partially purified enzyme revealed thermostability at 70oC without any loss of enzyme activity up to 10 hours with a half-life of 150 min at 70oC which drastically reduced (8-fold) at 90 oC. The enzyme showed a Km and Vmax values of 0.597 mol.min-1 and 13825 mg.ml-1, respectively. The application potential of enzyme in leather processing industry was evaluated.
